Biological Background: AK2 Function and Localization

  • Adenylate kinase 2 (AK2) is a mitochondrial enzyme that catalyzes the reversible transfer of the terminal phosphate group between ATP and AMP, playing a central role in cellular energy homeostasis and adenine nucleotide metabolism.
  • AK2 is critical for the regulation of phosphate utilization and the AMP de novo biosynthesis pathways.
  • It plays a key role in hematopoiesis (blood cell formation).
  • The protein localizes to the mitochondrial intermembrane space.
  • AK2 is highly expressed in heart, liver, and kidney, with lower levels in brain, skeletal muscle, and skin. It is present in thrombocytes and all nucleated blood cells, but absent in erythrocytes (which lack mitochondria). In leukocytes, AK2 is the predominant adenylate kinase, as AK1 is largely absent.
  • Post-translational modifications include acetylation and phosphorylation; the protein contains disulfide bonds and nucleotide-binding domains.
  • Mutations in AK2 are associated with reticular dysgenesis, a severe form of severe combined immunodeficiency (SCID).
  • Alternative splicing yields multiple transcript variants.

Experimental Guidance and Technical Tips

  • For Western blot, a band near 26 kDa is expected for endogenous AK2; using knockout cell lysates as a negative control is recommended to confirm specificity.
  • For immunofluorescence/ICC, permeabilization is required to detect mitochondrial AK2; co-localization with mitochondrial markers may help verify localization.
  • The antibody cross-reacts with mouse and rat AK2; verify target sequence conservation in these species before use.
  • For ELISA, the immunogen peptide can serve as a positive control; validate assay conditions using recombinant AK2 protein.
