Biological Background: KLF6 Function and Localization
- Also known as Zf9, COPEB, CPBP, and BCD1, KLF6 is a zinc-finger transcription factor from the Krüppel-like factor family.
- Functions as a transcriptional activator that binds to GC-rich promoter elements, regulating genes involved in cell growth and differentiation.
- Contains three C-terminal C2H2-type zinc finger motifs responsible for DNA binding and transcriptional activity.
- Localizes to the nucleus where it interacts with target gene promoters.
- Widely expressed, with highest levels in placenta, spleen, thymus, prostate, testis, small intestine, and colon; lower expression in pancreas, lung, liver, heart, and skeletal muscle.
- Implicated in B-cell development and may act as a tumor suppressor in certain contexts.
- May undergo alternative splicing, producing isoforms with potentially distinct regulatory roles.
- Included in proteomics identification and serves as a reference proteome component.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 40–200 of human KLF6, the N-terminal region upstream of the zinc-finger domains; this sequence may be conserved in mouse, consistent with reported cross-reactivity.
- For Western blot, expect a band around 32 kDa; validate in positive control lysates such as placenta or spleen with appropriate molecular weight markers.
- In immunohistochemistry (paraffin-embedded sections), consider antigen retrieval optimization and validate in tissues known to express KLF6.
- For immunofluorescence or ICC, nuclear staining is expected; include DAPI counterstain to confirm localization.
- Titrate the antibody for each application and sample type using a dilution series to determine optimal working concentration.
- When using mouse models, perform parallel control experiments to confirm cross-reactivity and specificity.
