Biological Background: KHSRP Function and Localization
- Far upstream element-binding protein 2 (FUBP2), also known as KH type-splicing regulatory protein (KSRP) or p75, is a 73 kDa RNA-binding protein.
- Binds to the dendritic targeting element and may play a role in mRNA trafficking. Part of a ternary complex that binds to the downstream control sequence (DCS) of pre-mRNA and mediates exon inclusion in tissue-specific alternative splicing.
- Involved in degradation of inherently unstable mRNAs that contain AU-rich elements (AREs) in their 3'-UTR, possibly by recruiting degradation machinery to ARE-containing mRNAs.
- May interact with single-stranded DNA from the far-upstream element (FUSE) and potentially activate gene expression.
- Subcellular localization: Found in both the nucleus and cytoplasm, consistent with roles in mRNA processing and transport.
- Tissue specificity: Detected in neural and non-neural cell lines.
- Post-translational modifications and keywords: Classified as a phosphoprotein, also subject to acetylation, methylation, and ubiquitination. Contains isopeptide bonds and has been identified by proteomics.

- For Western blot (WB), the antibody is raised against the N-terminal region (aa 1-180) of human KHSRP, predicting a band at 73 kDa. Validate in human, mouse, or rat cell lysates.
- For immunohistochemistry (IHC-P), antigen retrieval conditions may need optimization. Use tissue sections from neural or non-neural origins, where KHSRP is expressed.
- In immunoprecipitation (IP), the antibody can capture endogenous KHSRP; include appropriate IgG controls and validate with known interacting partners.
