Biological Background: KHDRBS1/Sam68 Function and Localization
- KHDRBS1, also known as p62, p68, or Sam68 (Src-associated in mitosis 68 kDa protein), is a KH domain-containing RNA-binding protein that functions as a signal transduction adapter.
- Upon tyrosine phosphorylation by receptors such as T-cell, leptin, and insulin receptors, KHDRBS1 binds SH2 and SH3 domain-containing proteins to propagate downstream signaling.
- KHDRBS1 modulates alternative splicing of numerous targets: CD44 (PubMed:26080397), BCL2L1 and SMN1 (PubMed:17371836, PubMed:20186123), and NRXN1/NRXN3 (by similarity). Related references: PMID:26080397, PMID:17371836, PMID:20186123
- It has been implicated in regulating mRNA stability, translation, and nuclear export; however, its role in CTE-containing RNA export is debated (PubMed:22253824). Related references: PMID:22253824

- Subcellularly, KHDRBS1 localizes to the nucleus, cytoplasm, and membrane.
- Ubiquitously expressed across tissues; isoform 1 is lower in brain, skeletal muscle, and liver, while isoform 3 is enhanced in skeletal muscle and liver. Isoform 3, expressed in growth-arrested cells, inhibits S phase.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide mapping to amino acids 1–100 of human KHDRBS1, targeting the N-terminal region. This region is present in all isoforms, so the antibody may recognize multiple variants; consider verifying isoform-specific reactivity.
- For Western blotting, a band corresponding to ~48 kDa is expected; however, due to post-translational modifications, multiple bands or shifts may be observed. Optimize loading and detection conditions.
- The antibody is validated for use in human, mouse, and rat samples. For applications not listed (e.g., IP, ChIP), validate in the relevant sample system.
- For IHC-P and IF/ICC, appropriate fixation and antigen retrieval methods should be optimized. Positive and negative controls are recommended.
- ELISA may be used for quantitative detection; pairing with a capture antibody can improve sensitivity in sandwich formats.
