Biological Background: STAT2 Function and Localization
- STAT2 (Signal transducer and activator of transcription 2), also known as p113, is a key mediator of type I interferon (IFN-alpha/beta) signaling. Upon receptor binding, TYK2 and JAK1 phosphorylate STAT2, allowing dimerization with STAT1 and association with IRF9 to form the ISGF3 complex, which activates interferon-stimulated genes (ISGs) via ISRE elements. Related references: PMID:23391734, PMID:9020188
- Beyond its transcriptional activator role, STAT2 also exerts negative feedback on type I IFN signaling by recruiting USP18 to the IFNAR2 receptor subunit, mitigating the antiviral response. Related references: PMID:28165510
- STAT2 is involved in regulating mitochondrial fission by modulating DNM1L phosphorylation at Ser-616 (activation) and Ser-637 (inactivation), linking innate immunity to mitochondrial dynamics. Related references: PMID:23391734, PMID:26122121
- The protein contains an SH2 domain and undergoes phosphorylation; it is a phosphoprotein and subject to ubiquitin-like conjugation (Ubl conjugation) as highlighted by UniProt keywords.
- STAT2 localizes to both the cytoplasm and the nucleus, with translocation occurring upon activation. Keywords include DNA-binding, transcription regulation, and antiviral defense.
- Alternative splicing of STAT2 generates multiple isoforms, and disease-associated variants have been reported (UniProt keyword: Disease variant).
- The protein interacts with host machinery during viral infection, indicated by the "Host-virus interaction" keyword; it is also identified in proteomics studies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 550–706 of human STAT2 (NP_005410.1). Verify that your target isoform or experimental conditions preserve this epitope region.
- For Western blot, anticipate a band near 98 kDa. Consider using interferon-alpha-stimulated cell lysates as positive controls to enhance detection.
- For ELISA, the antibody may be used for capture or detection; validate pairing and cross-reactivity with related STAT family proteins in your system.
