Biological Background: FOXK1 Function and Localization

  • Forkhead box protein K1 (FOXK1), also known as myocyte nuclear factor (MNF), is a transcriptional regulator with a predicted molecular weight of ~75 kDa.
  • Transcriptional regulator that binds the forkhead DNA sequence motif (5'-GTAAACA-3') and can act as either an activator or repressor depending on the context. Related references: PMID:17670796
  • Key regulator of aerobic glycolysis: together with FOXK2, promotes expression of glycolytic enzymes (HK2, PFK, PKLR, LDH) and suppresses mitochondrial pyruvate oxidation via PDK1/4 upregulation.
  • Negative regulator of autophagy in skeletal muscle: in response to starvation, represses autophagy genes to limit proteolysis.
  • Regulates myogenic progenitor cells: promotes proliferation by repressing FOXO4 and inhibits differentiation by blocking MEFC activity.
