Biological Background: XPNPEP3 Function and Localization

  • Catalyzes the removal of a penultimate prolyl residue from the N-termini of peptides, such as Leu-Pro-Ala; also shows low activity towards peptides with Ala or Ser at the P1 position. Related references: PMID:25609706 PMID:28476889
  • Promotes TNFRSF1B-mediated phosphorylation of MAPK8/JNK1 and MAPK9/JNK2, acting as an adapter protein independent of its catalytic activity, and may inhibit TNF-TNFRSF1B-induced apoptosis.
  • Two isoforms exist: isoform 1 is more abundant and widely expressed compared to isoform 2.
  • Subcellular localization is primarily mitochondrial, but also cytoplasmic.
  • Requires manganese as a cofactor and contains an N-terminal transit peptide for mitochondrial import.
  • Mutations in XPNPEP3 cause nephronophthisis, an autosomal recessive cystic kidney disease.
  • The molecular weight of the mature protein is approximately 57 kDa.
