Biological Background: TFEB Function and Localization

  • TFEB (Transcription factor EB), also known as Class E basic helix-loop-helix protein 35 (bHLHe35), belongs to the MiT/TFE subfamily of basic helix-loop-helix leucine zipper (bHLH-Zip) transcription factors, which includes MITF and TFE3.
  • It functions as a master regulator of lysosomal biogenesis and autophagy by binding to coordinated lysosomal expression and regulation (CLEAR) motifs (5′-GTCACGTGAC-3′) in the promoters of lysosomal and autophagic genes. Related references: PMID:19556463, PMID:22692423
  • TFEB activity is tightly controlled by nutrient-sensitive phosphorylation: under fed conditions, mTOR-mediated phosphorylation at multiple serine residues retains TFEB in the cytosol; starvation or lysosomal stress inhibits mTOR, leading to rapid dephosphorylation and nuclear translocation.   • The immunogen corresponds to amino acids 50–173 of human TFEB (NP_009093.1). This region lies within the N-terminal transcriptional activation domain, which is relatively conserved across human, mouse, and rat, supporting the reported cross-reactivity. For species not listed, sequence alignment is recommended.
  • For Western blot, TFEB may appear as a doublet between 44 and 53 kDa due to differential phosphorylation or alternative splicing; using fresh phosphatase inhibitors in lysis buffer can help preserve phospho-isoforms. The antibody has been validated for knockdown specificity (KD Validated).
  • In immunohistochemistry (IHC-P), antigen retrieval conditions should be optimized for your tissue type. Nuclear and cytoplasmic staining may be observed depending on the activation state of TFEB.
