Biological Background: Sharpin Function and Localization

  • Sharpin (Shank-associated RH domain-interacting protein), also known as SIPL1 (Shank-interacting protein-like 1), is a component of the linear ubiquitin chain assembly complex (LUBAC). LUBAC conjugates linear polyubiquitin chains in a head-to-tail manner to substrates, playing a key role in NF-kappa-B activation and inflammation regulation.

  • Through LUBAC, Sharpin mediates linear ubiquitination of IKBKG (NEMO) and RIPK1, which is essential for canonical NF-kappa-B and JNK signaling pathways. This modification interferes with TNF-induced cell death, thereby preventing inflammation.

  • For Western blot (WB), a band of approximately 40 kDa is expected. Use positive control lysates such as HeLa or HEK293, which express Sharpin.
  • In immunofluorescence/immunocytochemistry (IF/ICC), expect mainly cytoplasmic and possibly synaptic staining patterns. Optimize fixation and permeabilization for the cell type of interest.
  • ELISA application is possible; perform appropriate antibody titration and use recombinant Sharpin protein as standard.
  • As Sharpin is involved in TNF signaling, stimulation with TNF-alpha may affect its localization or complex formation, which could be probed using this antibody.
