Biological Background: PTBP1 Function and Localization

  • PTBP1, also known as hnRNP I, is a ubiquitously expressed RNA-binding protein involved in pre-mRNA splicing and alternative splicing regulation. It binds to polypyrimidine tracts of introns to modulate splice site selection.
  • It plays a key role in the regulation of tissue-specific alternative splicing events, such as activating exon skipping of its own pre-mRNA during muscle cell differentiation.
  • PTBP1 regulates splicing of several disease-relevant transcripts: it represses MAPT/Tau exon 10 inclusion, which is important in tauopathies. Related references: PMID:15009664
  • It promotes exon skipping of CFTR exon 9 by binding to the polypyrimidine-rich controlling element (PCE), antagonizing TIA1-mediated exon inclusion. Related references: PMID:14966131
  • In cancer metabolism, PTBP1 regulates pyruvate kinase PKM splicing by repressing exon 9 inclusion, leading to production of the PKM M2 isoform, which supports aerobic glycolysis. Related references: PMID:20010808
  • During picornavirus infection, PTBP1 binds to the viral internal ribosome entry site (IRES) and stimulates IRES-mediated translation, facilitating viral protein synthesis. Related references: PMID:21518806
  • Subcellular localization is predominantly nuclear, consistent with its role in pre-mRNA processing.
  • PTBP1 undergoes various post-translational modifications including acetylation, phosphorylation, and ubiquitination, which may regulate its activity and interactions.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human PTBP1. This region may be conserved across species, supporting the observed cross-reactivity with mouse and rat.
  • For Western blotting (WB), the expected molecular weight is ~60 kDa. Consider using appropriate loading controls and validate the band specificity in your target tissue or cell line.
  • When performing immunohistochemistry-paraffin (IHC-P), antigen retrieval conditions may need to be optimized for your specific sample type. Consider testing different retrieval buffers.
  • In immunofluorescence (IF/ICC), PTBP1 nuclear localization should be confirmed; nuclear counterstaining is recommended to verify localization.
  • For immunoprecipitation (IP), the monoclonal nature of this antibody provides high specificity. Titrate the antibody amount to achieve efficient capture without excessive background.
  • ELISA application may be used for quantitative detection; ensure coating conditions and detection system are compatible with the antibody format.
