Biological Background: mTOR Function and Localization
- mTOR (Mechanistic target of rapamycin), also known as FRAP, FRAP1, FRAP2, RAFT1, and RAPT1, is a 289 kDa serine/threonine protein kinase that functions as a central regulator of cellular metabolism, growth, and survival in response to hormones, growth factors, nutrients, energy, and stress signals. Related references: PMID:12087098 PMID:12150925 PMID:12150926
- mTOR forms two distinct multi-protein complexes, mTORC1 and mTORC2. mTORC1 is activated by nutrients and growth factors at the lysosome membrane, where it promotes anabolic processes such as protein, lipid, and nucleotide synthesis, while inhibiting catabolic pathways like autophagy. Related references: PMID:15268862 PMID:15467718 PMID:18497260 PMID:32561715
- mTORC1 directly phosphorylates key translational regulators, including EIF4EBP1 and RPS6KB1, to stimulate mRNA translation and ribosome biogenesis, thereby driving cell growth. Related references: PMID:24403073 PMID:29236692 PMID:12087098
- mTORC2 functions as a nutrient-insensitive complex that transduces growth factor signals, primarily phosphorylating and activating AGC kinases such as AKT and PKC, thereby regulating proliferation, cytoskeletal organization, and metabolism. Related references: PMID:15268862 PMID:15467718 PMID:24670654
- Subcellularly, mTOR is localized to various membranes, including lysosome, endoplasmic reticulum, Golgi apparatus, and cell membrane, as well as mitochondria, cytoplasm, and nucleus, which reflects its diverse signaling roles.
- mTOR is ubiquitously expressed with highest levels in testis, and is subject to multiple post-translational modifications, including acetylation, ubiquitination, and extensive phosphorylation.
- Dysregulation of mTOR signaling is associated with diseases such as epilepsy and intellectual disability, underscoring its critical role in cellular and organismal homeostasis.
Experimental Guidance and Technical Tips
- The immunogen used for this antibody corresponds to the N-terminal amino acids 1-300 of human mTOR, a region that may be suitable for detecting both mTORC1 and mTORC2 complex forms. Consider validating the antibody in your specific sample types and applications, as performance may vary depending on experimental conditions.
- This antibody has been validated for Western blot (WB), immunohistochemistry-paraffin (IHC-P), and ELISA. For WB, a high molecular weight band (~289 kDa) is expected; ensure appropriate gel conditions and transfer protocols for large proteins. For IHC-P, optimize antigen retrieval and antibody dilution to achieve specific staining.
- Cross-reactivity with mouse and rat mTOR has been confirmed; verify reactivity in your model system if using samples from other species.
