Biological Background: MAD1/MAD1L1 Function and Localization

  • MAD1L1 (also known as MAD1, TXBP181) encodes the mitotic spindle assembly checkpoint protein MAD1, a critical guardian of mitotic fidelity.
  • It forms a heterotetrameric complex with closed MAD2L1 (C-MAD2) at unattached kinetochores, promoting conversion of open MAD2L1 (O-MAD2) to C-MAD2 to propagate the “wait anaphase” signal. Related references: PMID:10049595 PMID:20133940 PMID:29162720
  • In hepatocellular carcinoma, MAD1 can sequester MAD2L1 in the cytoplasm, impairing the spindle assembly checkpoint and contributing to chromosomal instability.
  • The protein dynamically localizes to kinetochores, centrosomes, spindle poles, and the nuclear envelope during mitosis, and also shuttles through the cytoplasm.
  • MAD1 is subject to post-translational modifications, including phosphorylation and acetylation, which regulate its cell-cycle-dependent functions.
  • Tissue expression is notable in hepatocellular carcinomas and hepatoma cell lines, aligning with its oncogenic relevance.
  • Alternative splicing of MAD1L1 generates multiple isoforms, and mutations are associated with cancer-related checkpoint defects.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 351–718 of human MAD1, covering a large C-terminal region conserved across isoforms; this polyclonal antibody may therefore recognize multiple variants.
  • For Western blot, the expected molecular weight is approximately 83 kDa; use a 10% SDS-PAGE gel and ensure complete transfer for high-molecular-weight proteins.
  • ELISA reactivity should be validated under the specific assay conditions; consider checkerboard titrations to determine optimal coating antigen and antibody concentrations.
  • As with any polyclonal reagent, lot-to-lot consistency should be confirmed using a standardized cell lysate or recombinant protein.
