Biological Background: LXR beta/NER Function and Localization
- LXR beta (NR1H2) is a member of the nuclear receptor superfamily and serves as an oxysterol receptor, exhibiting ligand-dependent transcriptional activation. Related references: PMID:25661920
- It binds to specific DNA sequences known as direct repeat 4 (DR-4) elements, characterized by the consensus half-site AGGTCA separated by 4 nucleotides.
- LXR beta plays a key role in cholesterol metabolism by promoting MYLIP-mediated ubiquitination and subsequent degradation of LDLR, VLDLR, and LRP8 receptors.
- In hepatocytes, it induces LPCAT3-dependent phospholipid remodeling in the ER membrane, which drives SREBF1 processing, lipogenesis, and VLDL particle formation.
- The receptor also counteracts lipid-induced ER stress and inflammation, likely by modulating SRC kinase membrane compartmentalization.
