Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–100) of human LC3B, which is present in both the unconjugated (LC3B-I) and lipidated (LC3B-II) forms. Consider this design when interpreting results across different autophagic states.
- For Western blotting, a starting dilution of 1:500–1:2000 is often suitable; however, optimization in your experimental system is recommended. Expect to observe two bands corresponding to LC3B-I and LC3B-II, with LC3B-II migrating slightly faster.
- In immunofluorescence (IF-P) and immunohistochemistry (IHC-P), include appropriate controls (e.g., autophagy induction) and validate signal specificity with knockdown or knockout samples if available.
- Since the antibody shows cross-reactivity with mouse and rat LC3B, confirm sequence homology if using these species, and consider performing a pilot study to assess performance in non-human samples.
