Biological Background: ISG15 Function and Localization

  • ISG15 (interferon-stimulated gene 15) is a 15-17 kDa ubiquitin-like protein, also known as G1P2, IP17, and hUCRP, encoded by the ISG15 gene on human chromosome 1.

  • ISGylation promotes oligomerization of the viral sensor IFIH1/MDA5, activating innate immunity against coronaviruses, flaviviruses, and picornaviruses. Related references: PMID:33727702
  • Free, unconjugated ISG15 is secreted and acts as a cytokine, inducing IFN-γ production and NK cell proliferation, and serving as a chemotactic factor for neutrophils. Related references: PMID:29100055
  • Subcellularly, ISG15 is found in the cytoplasm and can be secreted into the extracellular space.
  • It exhibits broad antiviral activity against both DNA and RNA viruses, including influenza A, HIV-1, and Ebola virus, by disrupting viral budding and replication.

  • The immunogen corresponds to full-length human ISG15 (amino acids 1-165), suggesting antibody recognition of the native protein. Confirm specificity in your sample matrix.
  • For western blot, the expected band is at ~18 kDa under reducing conditions. Use appropriate positive controls (e.g., interferon-treated cells) to verify detection.
  • Cross-reactivity with mouse and rat ISG15 is reported; however, validate reactivity in your specific experimental system, as species homology may affect signal intensity.
