Biological Background: ISG15 Function and Localization
- ISG15 (Interferon-stimulated gene 15) is a ubiquitin-like protein also known as G1P2, IP17, or hUCRP, encoded by the ISG15 gene.
- It plays a key role in innate immunity, acting either via conjugation to target proteins (ISGylation) or as a free secreted cytokine.
Related references: PMID:27564865 PMID:39465252 - ISGylation involves E1, E2, and E3 enzymes and targets proteins such as IFIT1, MX1, and the viral sensor IFIH1/MDA5, promoting antiviral signaling.
Related references: PMID:33727702 - Exhibits broad antiviral activity against DNA and RNA viruses, including influenza A, HIV-1, and Ebola, by disrupting viral budding or protein function.
- The secreted form of ISG15 acts as a cytokine, inducing natural killer cell proliferation, neutrophil chemotaxis, and IFN-gamma production, essential for antimycobacterial immunity.
Related references: PMID:29100055 - ISG15 is localized in the cytoplasm and also secreted; expression is found in lymphoid cells, muscle, epithelia, and neurons, and is upregulated in certain cancers.
- Post-translational modifications include disulfide bond formation and S-nitrosylation; ISG15 is part of the ubiquitin-like conjugation pathway.
Experimental Guidance and Technical Tips
- This antibody was generated against a recombinant protein covering amino acids 1–165 of human ISG15, corresponding to the full-length canonical isoform; it is expected to detect the ~18 kDa protein in Western blotting.
- For WB, consider using standard lysates from interferon-stimulated human cells to confirm ISG15 upregulation; a single band near 18 kDa should be observed.
- Cross‑reactivity is reported for human samples only; if extending to other species, sequence homology should be checked and validation performed.
- As a polyclonal antibody, lot‑to‑lot consistency should be verified for critical quantitative assays.
