Biological Background: Endoplasmin Function and Localization

  • HSP90B1, also known as Endoplasmin, GRP-94, or gp96, is a 92-kDa heat shock protein of the HSP90 family.
  • It functions as an ATP-dependent molecular chaperone in the endoplasmic reticulum, regulating protein folding and transport, and together with MESD, modulates the Wnt pathway by promoting LRP6 folding. Related references: PMID:23572575, PMID:39509507
  • In association with CNPY3, it is required for proper folding of Toll-like receptors and promotes TLR4 trafficking to the cell surface. Related references: PMID:11584270
  • It may participate in unfolding of cytosolic leaderless cargos, such as interleukin-1, to facilitate their TMED10-mediated translocation into the ERGIC for unconventional secretion. Related references: PMID:32272059
  • Endoplasmin is localized in the lumen of the endoplasmic reticulum, sarcoplasmic reticulum, and melanosomes.
  • Post-translational modifications include acetylation, phosphorylation, hydroxylation, glycosylation, and the presence of a disulfide bond; it binds ATP and calcium.
  • Designated as tumor rejection antigen 1 (gp96), it is implicated in immune responses against tumors and has been studied in cancer vaccine contexts.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 616–715) of human HSP90B1, which is highly conserved across human, mouse, and rat, explaining the broad species reactivity. When applying this antibody to other species, verify sequence homology.
  • For Western blotting, expect a band near 92 kDa under reducing conditions; validate in appropriate positive control lysates and consider using loading controls and molecular weight markers.
  • In IHC-P, optimize antigen retrieval conditions and antibody dilution for the target tissue; Endoplasmin is abundant in ER-rich secretory cells, such as hepatocytes and plasma cells.
  • For ELISA, this antibody can serve as a capture or detection reagent; pairing with an antibody against a distinct epitope may improve assay sensitivity and specificity.
