Biological Background: GM130 Function and Localization
- GM130 (GOLGA2) is a 130 kDa cis-Golgi matrix protein also known as golgin-95, belonging to the golgin family of coiled-coil proteins.
- Acts as a peripheral membrane skeleton maintaining the structure of the Golgi apparatus and facilitating vesicle tethering to the Golgi membrane. Related references: PMID:16489344
- Together with p115/USO1 and STX5, mediates vesicle tethering and fusion at the cis-Golgi network, essential for the stacked and interconnected Golgi architecture.
- Required for normal protein transport from the endoplasmic reticulum to the Golgi and cell membrane (By similarity).
- Central to mitotic Golgi disassembly: phosphorylation at Ser-37 by CDK1 at mitosis onset disrupts p115/USO1 interaction, blocking COPI vesicle tethering and halting Golgi transport.
- Plays a critical role in spindle pole assembly and centrosome organization by activating TPX2 through importin-alpha sequestration, thereby nucleating microtubules around the Golgi; also regulates meiotic spindle assembly. Related references: PMID:19242490, PMID:26165940
