Biological Background: Glucose-6-phosphate isomerase Function and Localization

  • Catalyzes the conversion of alpha-D-glucose-6-phosphate to beta-D-fructose-6-phosphate, the second step in glycolysis, and the reverse reaction in gluconeogenesis. Related references: PMID:28803808
  • Also displays C2-epimerase and anomerase activities, interconverting D-glucose-6-phosphate and D-mannose-6-phosphate, and alpha/beta anomeric forms.
  • Functions as autocrine motility factor (AMF), a secreted angiogenic factor that enhances endothelial cell motility. Related references: PMID:11437381
  • Acts as neuroleukin (NLK), a neurotrophic factor supporting the survival of spinal and sensory neurons. Related references: PMID:11004567 PMID:3352745
  • Released by lectin-stimulated T-cells to induce immunoglobulin secretion. Related references: PMID:11004567 PMID:3352745
  • Subcellular localization: predominantly cytoplasmic, but also secreted.
  • Post-translational modifications include acetylation, hydroxylation, phosphorylation, and ubiquitination.
  • Associated with hereditary hemolytic anemia when mutated (keywords: disease variant).

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 1-100 of human GPI; consider validating reactivity in your human sample system before large-scale experiments.
  • For WB, use whole-cell lysates from human cell lines and expect a band near 63 kDa; validate the knockdown efficiency with appropriate controls.
  • For ELISA, optimize antigen coating and antibody dilution to ensure specificity; consider paired detection reagents for quantitative assays.
  • Since the antibody is KD validated, confirm gene silencing in your experimental setup and include scrambled shRNA/siRNA controls.
