- Fibroblast growth factor receptor 3 (FGFR3), also designated CD333, JTK4, and ACH, is a member of the immunoglobulin superfamily of tyrosine kinase receptors. Multiple isoforms are generated through alternative splicing, including a secreted isoform (isoform 3) that retains ligand-binding ability.
- FGFR3 is a cell-surface receptor that binds fibroblast growth factors, triggering intracellular signaling cascades that regulate cell proliferation, differentiation, and apoptosis. It is essential for normal chondrocyte differentiation and skeletal development, and also modulates osteogenesis and postnatal bone mineralization.
- Ligand binding activates key pathways: phosphorylation of PLCG1 produces second messengers; FRS2 phosphorylation recruits GRB2/SOS1 leading to RAS-MAPK (ERK1/2) activation, and also engages the AKT1 signaling pathway. - Tissue expression includes brain, kidney, testis, and fetal tissues such as lung, small intestine, and kidney. Isoform 2 is detected in epithelial cells, while isoform 1 is found in fibroblastic cells.
- Dysregulation of FGFR3, often due to activating mutations, is implicated in skeletal dysplasias (e.g., achondroplasia, thanatophoric dysplasia), craniosynostosis, and multiple cancers. Constitutively active FGFR3 promotes STAT1, STAT5, and PTPN11/SHP2 signaling, contributing to oncogenesis.

- The immunogen spans the extracellular region (amino acids 23–375), which is present in all membrane-bound and secreted isoforms. Verify detection of desired isoforms in your sample matrix by consulting the literature and using isoform-specific controls.
- In Western blotting, expect multiple bands between 75 and 88 kDa due to differential glycosylation and isoform expression. Optimize loading controls and consider using deglycosylation enzymes to confirm specificity.
- For flow cytometry, use non-permeabilized cells to target surface FGFR3. Include a relevant isotype control and validate with FGFR3-overexpressing or siRNA-knockdown cell lines.
- In ELISA, the secreted isoform 3 may compete for capture or detection antibodies. Pre-clear culture supernatants or use monoclonal/polyclonal pairs designed against non-overlapping epitopes.
