Biological Background: EGFR Function and Localization
- EGFR is a receptor tyrosine kinase that binds EGF family ligands and activates multiple downstream signaling cascades, including RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, STAT, and NF-kappa-B modules, to convert extracellular cues into cellular responses like proliferation, survival, and migration. - For Western blotting, multiple bands may be observed corresponding to full-length EGFR (~134 kDa) and proteolytic fragments (e.g., 44, 69, 77 kDa); these are consistent with reported EGFR processing.
- In immunofluorescence/ICC, expect plasma membrane and endosomal staining; permeabilization may be required to detect intracellular pools.
