Biological Background: DEC1 Function and Localization

  • BHLHE40 (Class E basic helix-loop-helix protein 40), commonly known as DEC1, SHARP-2, or STRA13, is a transcriptional repressor with a central role in circadian rhythm regulation.
  • It acts as the negative limb of the DEC autoregulatory feedback loop, repressing the CLOCK-BMAL1 heterodimer by competing for E-box elements (5'-CACGTG-3') within target gene promoters, thereby controlling clock and clock-controlled genes. Related references: PMID:12397359 PMID:18411297 PMID:14672706 PMID:15560782
  • The DEC loop is interlocked with the PER/CRY loop: DEC1 represses PER1/2 expression and is itself repressed by PER1/2 and CRY1/2. Related references: PMID:15193144
  • DEC1 may regulate chondrocyte differentiation through the cAMP signaling pathway. Related references: PMID:19786558
  • It functions as a corepressor of RXR and RXR-LXR heterodimers, attenuating ligand-induced transactivation of RXRA and NR1H3/LXRA. Related references: PMID:19786558
  • Drives the circadian rhythm of blood pressure via transcriptional repression of ATP1B1 in the cardiovascular system. Related references: PMID:30012868
  • Subcellular localization is primarily nuclear and cytoplasmic.
  • Expressed in cartilage, spleen, intestine, lung, and to a lesser extent in heart, brain, liver, muscle, and stomach.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 121–300 of human DEC1 (NP_003661.1), likely spanning functionally important regions; consider epitope accessibility when designing assays.
  • For Western blot, a band near 46 kDa is expected; UniProt lists a full-length molecular weight of ~45.5 kDa, and slight shifts may occur due to phosphorylation or other modifications.
  • This antibody is recommended for WB and ELISA applications; if using in other techniques, validate performance in the relevant sample system.
  • Reported cross-reactivity is for human samples; for non-human species, sequence homology analysis is advised prior to use.
