Biological Background: CREB-binding protein (CREBBP) Function and Localization
- CREBBP (also known as CBP, KAT3A) is a histone lysine acetyltransferase and transcriptional coactivator encoded by the CREBBP gene.
- Mediates acetylation of histone H3 at Lys18 and Lys27 (H3K18ac and H3K27ac), creating specific tags for transcriptional activation. Related references: PMID:21131905, PMID:24616510
- Acetylates non-histone proteins such as PCNA (promoting its removal during nucleotide excision repair) and POLR1E (decreasing RNA polymerase I association with rDNA). Related references: PMID:24939902, PMID:24207024
- Acts as a circadian transcriptional coactivator, enhancing the activity of NPAS2-BMAL1 and CLOCK-BMAL1 heterodimers. Related references: PMID:14645221
- In addition to acetyl-CoA, can use lactoyl-CoA to mediate protein lactylation, as seen with MRE11 in response to DNA damage. Related references: PMID:38128537
- Localizes to both the cytoplasm and nucleus, consistent with its diverse roles in transcription and repair.
- Contains multiple conserved domains including a bromodomain, zinc fingers, and a histone acetyltransferase (HAT) domain, characteristic of the p300/CBP family.
Experimental Guidance and Technical Tips
- For WB, use a low-percentage gel (e.g., 6-8%) and optimized transfer for high-molecular-weight protein (265 kDa).
- For IF/ICC, fix cells with 4% PFA and permeabilize with 0.1% Triton X-100; consider blocking with 5% BSA.
- For IHC-P, perform heat-mediated antigen retrieval in citrate buffer (pH 6.0); test antibody dilution series.
- Validate reactivity in the appropriate cell lines or tissues; the antibody is reported to cross-react with human, mouse, and rat.
- The immunogen (amino acids 2343–2442) targets a C-terminal region; be aware of potential isoforms.
