Biological Background: Cyclin-T1 Function and Localization
- Cyclin-T1 is the regulatory subunit of the positive transcription elongation factor b (P-TEFb) complex, which consists of CDK9 and cyclin T1. P-TEFb phosphorylates the C-terminal domain (CTD) of RNA polymerase II to promote productive transcriptional elongation (PubMed:16109376, PubMed:16109377, PubMed:30134174). Related references: PMID:16109376, PMID:16109377, PMID:30134174
- Cyclin-T1 activates CDK9 by mediating liquid-liquid phase separation (LLPS), which enhances P-TEFb binding to the CTD of RNA Pol II (PubMed:29849146, PubMed:35393539). Related references: PMID:29849146, PMID:35393539
- During HIV-1 or SIV infection, Cyclin-T1 binds the viral transactivator Tat, increasing its affinity for TAR RNA and serving as an essential cofactor for viral gene transcription.
- Cyclin-T1 is ubiquitously expressed and localizes to the nucleus.
- The protein undergoes various post-translational modifications, including phosphorylation, acetylation, and ADP-ribosylation, which regulate its activity.
- Alternative splicing of the CCNT1 gene generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide within amino acids 621–720 of human Cyclin T1, targeting the C‑terminal region.
- This antibody has been shown to detect human Cyclin T1 in Western blot, immunoprecipitation, immunofluorescence, and ELISA applications; consider optimizing for your specific experimental conditions.
- Validate the antibody in your relevant sample system and include appropriate controls.
