- It functions as an ecotropic retroviral leukemia receptor, serving as a cell surface receptor for ecotropic murine leukemia viruses (hence the alternate names ERR and REC1L).
- The protein is a member of the solute carrier family 7 (SLC7) and contains multiple transmembrane domains, consistent with its role in membrane transport.
- CAT1 is ubiquitously expressed across human tissues, ensuring systemic availability of essential cationic amino acids.
- Subcellularly, it localizes to the cell membrane, where it facilitates the influx of cationic amino acids from the extracellular environment into the cytoplasm.
- The protein is glycosylated and has a molecular weight of approximately 68 kDa as determined by SDS-PAGE (calculated mass: 67.6 kDa).
Experimental Guidance and Technical Tips
- For Western blot, consider using a dilution range of 1:500–1:2000; a specific band around 68 kDa is expected based on the predicted molecular weight and endogenous glycosylation.
- The antibody may also detect mouse CAT1 due to reported cross-reactivity; suitable positive controls include human and mouse cell lysates.
- Antigen retrieval is not required for Western blotting; standard SDS-PAGE and transfer conditions are typically adequate.
- In ELISA applications, coat the synthetic peptide immunogen or recombinant protein and validate the antibody’s binding affinity under your specific assay conditions.
