Biological Background: BUB1 Function and Localization
- Mitotic checkpoint serine/threonine-protein kinase, also known as BUB1A, encoded by the BUB1 gene. Essential for the spindle-assembly checkpoint (SAC) and correct chromosome alignment during mitosis.
- Required for the kinetochore localization of multiple checkpoint proteins including CENPF, BUB1B, CENPE, MAD2L1, PLK1, and AURKB, thereby ensuring robust SAC signaling.
- Forms a complex with BUB3 that inhibits the anaphase-promoting complex/cyclosome (APC/C) by phosphorylating its activator CDC20; also phosphorylates MAD1L1 and BUB3.
- Promotes centromeric localization of TOP2A, contributing to proper chromosome segregation. Related references: PMID:35044816
- Plays a critical role in defining the localization of SGO1, thereby affecting sister chromatid cohesion and chromosome segregation.
- Acts as a substrate for APC/C-Cdh1 and mediates cell death in response to chromosome missegregation, suppressing spontaneous tumorigenesis.
- Localizes to the nucleus, concentrated at the centromere and kinetochore on chromosomes during mitosis.
- Highly expressed in testis and thymus; lower expression in colon, spleen, lung, and small intestine. Associated with tissues having a high mitotic index.
