Biological Background: ATG7 Function and Localization
- Ubiquitin-like modifier-activating enzyme ATG7 is a key E1-like enzyme that initiates the two ubiquitin-like conjugation systems essential for autophagy and cytoplasm-to-vacuole transport (Cvt).
- It activates ATG12 for conjugation with ATG5, and also activates the ATG8 family proteins (including LC3) for conjugation with phosphatidylethanolamine, a critical step in autophagosome membrane elongation.
- ATG7 facilitates the lipidation of LC3-I to form LC3-II, which associates with autophagosomal membranes. This process is crucial for autophagosome formation and maturation. Related references: PMID:34161705
- Beyond canonical autophagy, ATG7 is required for selective mitophagy, helping maintain mitochondrial homeostasis and prevent excessive ROS production.
- It modulates p53/TP53 activity, thereby influencing cell cycle progression and survival under metabolic stress, and plays roles in axonal homeostasis, hematopoiesis, and adipose differentiation.
- ATG7 localizes predominantly to the cytoplasm and preautophagosomal structures, consistent with its function in autophagosome biogenesis.
- The protein is widely expressed, with highest levels in kidney, liver, lymph nodes, and bone marrow. Post-translational modifications include acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-100) of human ATG7. Consider epitope accessibility when designing experiments.
- For Western blot, the expected band for ATG7 is approximately 78 kDa based on the UniProt molecular weight; validate in your sample system using appropriate positive and negative controls.
- This antibody is suitable for WB, IHC-P, IP, and ELISA per the supplier. Optimize dilutions and conditions for each application and sample type.
- Cross-reactivity is reported for human, mouse, and rat. If using other species, verify sequence homology in the immunogen region.
