Biological Background: ADAR1 Function and Localization

  • ADAR1 (also known as DRADA, p136, IFI-4) is a double-stranded RNA-specific adenosine deaminase that catalyzes the conversion of adenosine to inosine in dsRNA (A-to-I RNA editing). This post-transcriptional modification can alter codon recognition, splicing, RNA stability, and structure.

  • The protein plays a critical role in both cellular and viral RNA metabolism. Cellular substrates include neurotransmitter receptors (GRIA2, HTR2C, GABRA3) and the bladder cancer-associated protein BLCAP, where editing modifies amino acid sequences and functional properties.

  • ADAR1 exhibits dual roles in viral infection: it can exert antiviral effects (e.g., inhibiting HCV replication via hyper-editing) or proviral effects (e.g., enhancing HDV, MV, VSV, and HIV-1 replication).   • The immunogen corresponds to amino acids 610–810 of human ADAR1, targeting the C-terminal region. For Western blotting, the predicted molecular weight is ~136 kDa, but isoform-specific bands may be observed; validate using known positive controls.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider optimizing antigen retrieval and antibody dilution using tissues with known high expression (e.g., brain or lung). Co-staining with organelle markers may help confirm cytoplasmic or nuclear localization.
  • In ELISA applications, pair with a suitable capture or detection antibody and validate in the relevant sample matrix to ensure specificity for ADAR1.
