Biological Background: ACSS2 Function and Localization
- ACSS2, also known as acetyl-CoA synthetase 1 (AceCS1), is a cytoplasmic enzyme that catalyzes acetyl-CoA production from acetate, with acetate being the preferred substrate over propionate. Related references: PMID:10843999, PMID:28003429, PMID:38369012
- It does not utilize butyrate or crotonate as substrates. Related references: PMID:38369012
- Under glucose deprivation, AMPK phosphorylates ACSS2, leading to nuclear translocation where it interacts with TFEB to locally supply acetyl-CoA for histone acetylation at lysosomal and autophagy gene promoters, promoting tumor survival. Related references: PMID:28552616
- ACSS2 also acts as a chromatin-bound coactivator for neuronal gene expression, critical for memory consolidation, by maintaining acetyl-CoA for histone acetylation at memory gene loci (By similarity).
- It catalyzes the conversion of lactate into lactoyl-CoA, expanding its metabolic roles. Related references: PMID:39561764
- The protein localizes to both the cytoplasm and nucleus, reflecting its dual metabolic and transcriptional regulatory functions.
- Post-translational modifications include acetylation and phosphorylation, and the protein is annotated with keywords such as Activator, ATP-binding, Ligase, Lipid metabolism, Phosphoprotein, and Transcription regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an N-terminal peptide (aa 50-150) of human ACSS2; this region is conserved across species, supporting the observed human/mouse/rat cross-reactivity.
- In Western blotting, endogenous ACSS2 typically migrates at ~79-80 kDa; verify band specificity using appropriate controls (e.g., knockdown or knockout samples).
- For ELISA applications, pair with a suitable detection antibody and validate using recombinant ACSS2 protein or cell lysates from known positive samples.
- The antibody's knock-down validation status indicates suitability for gene silencing confirmation studies; however, researchers should perform independent validation in their experimental context.
