Biological Background: KCNU1 Function and Localization
- KCNU1 (Potassium channel subfamily U member 1), also known as Slo3, KCa5, is a testis-specific calcium-activated potassium channel encoded by the KCNU1 gene (KCNMA3, KCNMC1, SLO3).
- It is activated by both intracellular pH and membrane voltage, mediating potassium efflux and representing the primary spermatozoan K+ current (PubMed:23129643, PubMed:24670955, PubMed:36649421). Related references: PMID:23129643 PMID:24670955 PMID:36649421
- KCNU1 underlies the pH-triggered membrane hyperpolarization critical for sperm capacitation as sperm encounter the alkaline environment of the female reproductive tract, and it is essential for male fertility (PubMed:34980136, PubMed:35551387, PubMed:36649421). Related references: PMID:34980136 PMID:35551387 PMID:36649421
- It localizes to the cell membrane and is enriched in the flagellum membrane of sperm.
- Mutations in KCNU1 are associated with spermatogenic failure (SPGF79), highlighting its role in reproductive health.
- Predicted to contain multiple transmembrane helices and a voltage-gated potassium channel domain.
- Exclusively expressed in the testis, consistent with its specialized reproductive function.
Experimental Guidance and Technical Tips
- For western blot analysis, a band around 130 kDa is expected; testis or sperm lysates may serve as positive controls.
- In ELISA applications, consider using recombinant KCNU1 protein or peptide as the coating antigen to capture the antibody.
- The antibody shows cross-reactivity with rat, suggesting conserved epitopes; validate performance in rat testis samples if required.
- As a polyclonal antibody, it may provide strong signal but could also produce background; optimize blocking and washing conditions.
- The immunogen corresponds to a C-terminal region (aa 920-1149); verify detection of full-length protein in native samples.
