Biological Background: ITPK1 Function and Localization

  • ITPK1 is a human kinase/phosphatase/isomerase belonging to the inositol phosphate metabolism pathway, with alternative splice variants reported.
  • ITPK1 phosphorylates Ins(3,4,5,6)P4 at position 1 and Ins(1,3,4)P3 at positions 5 and 6, playing a key role in inositol phosphate synthesis. Related references: PMID:11042108 PMID:8662638
  • The protein also acts as an inositol polyphosphate phosphatase, dephosphorylating Ins(1,3,4,5)P4 and Ins(1,3,4,6)P4 to Ins(1,3,4)P3. Related references: PMID:11909533 PMID:17616525
  • ITPK1 may interconvert Ins(1,3,4,5)P4 and Ins(1,3,4,6)P4 in the presence of ADP and magnesium. Related references: PMID:11909533
  • It modulates TNF-induced apoptosis by interfering with the activation of TNFRSF1A-associated death domain. Related references: PMID:11909533 PMID:12925536 PMID:17616525
  • ITPK1 is essential for MLKL-mediated necroptosis by producing highly phosphorylated inositol phosphates that activate MLKL, leading to membrane localization. Related references: PMID:17616525
  • ITPK1 is expressed in multiple tissues, with highest levels in brain, and is also found in heart, skeletal muscle, kidney, pancreas, liver, placenta, and lung.
  • The protein is subject to acetylation and phosphorylation and requires magnesium as a cofactor.

Experimental Guidance and Technical Tips

  • The immunogen (recombinant fusion protein corresponding to amino acids 185-414) covers the C-terminal region of human ITPK1; this region may confer target specificity.
  • For Western blotting, use appropriate positive control lysates from human, mouse, or rat tissues expressing ITPK1 (e.g., brain or heart) and optimize antibody dilution.
  • For ELISA, coat with immunogen or target protein and validate under standard assay conditions.
  • Note that the predicted molecular weight is ~46 kDa, but actual migration may vary depending on post-translational modifications and gel conditions.
