Biological Background: ISG15 Function and Localization
- ISG15 is a ubiquitin-like protein that serves a crucial role in the innate immune response to viral infection, acting either via ISGylation of target proteins or as an unconjugated secreted signaling molecule. Related references: PMID:27564865 PMID:39465252
- ISGylation involves a cascade of E1, E2, and E3 enzymes that conjugate ISG15 to lysine residues on target proteins including IFIT1, MX1, IFIH1/MDA5, and IRF3, thereby modulating antiviral signaling. Related references: PMID:33727702
- The protein exhibits antiviral activity against both DNA and RNA viruses; it disrupts HIV-1 and Ebola virus budding and ISGylates influenza A virus NS1 protein to inhibit replication.
- Secreted ISG15 binds the ITGAL/ITGB2 integrin receptor to activate SRC family kinases, leading to secretion of IFNG and IL10, and plays an essential role in antimycobacterial immunity. Related references: PMID:29100055
- Subcellularly, ISG15 is localized in the cytoplasm and is also secreted into the extracellular space.
- Tissue expression is detected in lymphoid cells, striated and smooth muscle, epithelia, and neurons; overexpression correlates with advanced stages in pancreatic, endometrial, and bladder cancers.
- Keywords associated with ISG15 include Antiviral defense, Cytoplasm, Secreted, Innate immunity, and Ubl conjugation pathway.
Experimental Guidance and Technical Tips
- The immunogen corresponds to full-length human ISG15, so the polyclonal antibody may recognize multiple epitopes; consider using it in both native and denaturing conditions.
- For WB, use interferon-stimulated cell lysates (e.g., IFN-α/β treated HeLa or A549) as positive controls to induce ISG15 expression.
- For IHC-P, validate staining patterns with known tissue controls; ISG15 upregulation in certain cancers may serve as a relevant model.
- In ELISA, titrate the antibody with recombinant ISG15 protein to establish optimal working concentrations.
- Given cross-reactivity with rat, verify expected bands in rat samples if included in experiments.
