Biological Background: Integrin alpha-V Function and Localization
- Integrin alpha-V (ITGAV) is a cell surface receptor that heterodimerizes with various beta subunits (ITGB1, ITGB3, ITGB5, ITGB6, ITGB8) to form functional integrins.
- ITGAV-containing integrins recognize the RGD (Arg-Gly-Asp) motif present in extracellular matrix proteins such as vitronectin, fibronectin, fibrinogen, and laminin, mediating cell adhesion, migration, and signaling.
- ITGAV:ITGB3 binds to growth factors (FGF1, FGF2, IGF1, IGF2) and cytokines (NRG1, IL1B, fractalkine) to modulate critical signaling pathways including FGF, IGF, ERBB, IL1B, and CX3CR1-dependent signaling. - ITGAV:ITGB3 interacts with PLA2G2A via a distinct site, inducing conformational changes that enhance ligand binding. Related references: PMID:18635536 PMID:25398877
- ITGAV:ITGB3 and ITGAV:ITGB6 bind fibrillin-1 (FBN1) in an RGD-dependent manner, and ITGAV:ITGB6/ITGB8 activate TGF-beta-1 by releasing it from latency-associated peptide (LAP). Related references: PMID:12807887 PMID:17158881 PMID:15184403 PMID:22278742 PMID:28117447
- ITGAV is predominantly localized at the cell membrane and enriched at cell junctions and focal adhesions.
- ITGAV serves as a host cell receptor for several viruses, including Adenovirus, Coxsackievirus, Herpesvirus, Parechovirus, and West Nile virus, and is implicated in HIV-1 Tat-induced angiogenesis.
- Post-translational modifications include glycosylation, disulfide bond formation, and proteolytic cleavage, and ITGAV binds calcium and other metal ions essential for integrin function.
Experimental Guidance and Technical Tips
- The synthetic peptide immunogen corresponds to amino acids 949-1048 of human ITGAV, a region within the extracellular domain; this antigen design may be suitable for detecting both full-length and cleaved forms.
- For Western blot, the predicted molecular weight is ~116 kDa (full-length) or ~130 kDa (glycosylated); under reducing conditions, a heavy chain of ~125 kDa and a light chain of ~25 kDa may be observed after post-translational cleavage.
- For IHC-P, consider heat-induced antigen retrieval and verify fixation conditions; for flow cytometry (intracellular), permeabilization is required.
- The antibody cross-reacts with human, mouse, and rat; validate in the relevant sample system to confirm performance.
- These are general technical considerations; optimal experimental conditions should be determined empirically.
