Upon binding to IL4, the IL4R receptor dimerizes with common gamma chain to form the type 1 signaling complex on hematopoietic cells, or with IL13RA1 to form type 2 complex on nonhematopoietic cells, leading to JAK/STAT6 pathway activation (PubMed:10219247, PubMed:11526337, PubMed:18243101, PubMed:7721895).
