Biological Background: Interleukin-4 Function and Localization

• IL4 is a secreted cytokine primarily produced by mast cells, T-cells, eosinophils, and basophils that plays a central role in regulating antibody production, hematopoiesis, inflammation, and effector T-cell responses (PubMed:1993171, PubMed:3016727). Related references: PMID:1993171 PMID:3016727

• Upon binding to IL4, the IL4R receptor dimerizes with common gamma chain to form the type 1 signaling complex on hematopoietic cells, or with IL13RA1 to form type 2 complex on nonhematopoietic cells, leading to JAK/STAT6 pathway activation (PubMed:10219247, PubMed:11526337, PubMed:18243101, PubMed:7721895). • For WB, consider using reducing conditions; the predicted molecular weight is 17 kDa, but observe that glycosylation may alter migration.
• For ELISA, validate optimal coating concentrations and detection conditions in your sample matrix; since IL4 is secreted, appropriate sample preparation (e.g., cell culture supernatant or serum) is recommended.
