Biological Background: IL4 Function and Localization
- IL4, also known as B-cell stimulatory factor 1 (BSF-1) and Binetrakin, is a secreted cytokine encoded by the IL4 gene on chromosome 5 in humans.
- It is secreted primarily by activated T-cells, mast cells, eosinophils, and basophils.
- IL4 plays a central role in regulating antibody production, hematopoiesis, and inflammation, particularly by inducing class II MHC expression on resting B-cells and enhancing IgE and IgG1 secretion.

- The protein contains a signal peptide, disulfide bonds, and glycosylation sites, and is subject to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen fragment (aa 25-153) corresponds to the mature human IL4 protein and is designed to elicit reactivity against the native and recombinant cytokine.
- For Western blot, the theoretical molecular weight is ~17 kDa; however, glycosylation and disulfide bonding may result in a shifted band (15–20 kDa) under reducing conditions.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), we recommend optimizing antigen retrieval and antibody dilution in the relevant tissue context.
- For ELISA, this polyclonal antibody can be evaluated as a capture or detection reagent paired with a matched monoclonal antibody.
