Biological Background: Interleukin-4 Function and Localization
- Interleukin-4 (IL-4) is a secreted cytokine, also known as BSF-1, Binetrakin, Lymphocyte stimulatory factor 1, and Pitrakinra.
- Produced by mast cells, T-cells, eosinophils, and basophils, IL-4 is a central regulator of humoral and adaptive immunity, including antibody class switching and effector T-cell responses.
Related references: PMID:1993171, PMID:3016727 - IL-4 induces class II MHC expression on resting B-cells and enhances secretion and cell surface expression of IgE and IgG1.
Related references: PMID:1993171 - It also regulates expression of the low-affinity IgE receptor (CD23) on lymphocytes and monocytes.
Related references: PMID:2521231 - IL-4 positively regulates IL31RA in macrophages and stimulates autophagy in dendritic cells via mTORC1 inhibition and RUFY4 induction.
- Signaling occurs via two receptor complexes: type 1 (IL4R/IL2RG) on hematopoietic cells and type 2 (IL4R/IL13RA1) on non-hematopoietic cells, leading to JAK/STAT6 pathway activation.
Related references: PMID:10219247, PMID:11526337, PMID:18243101, PMID:7721895 - Beyond immune functions, IL-4 plays a role in normal brain physiology, including memory and learning (By similarity).
- The protein is secreted, glycosylated, and contains disulfide bonds; its molecular weight is approximately 17.5 kDa, with multiple isoforms from alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen was designed using a synthetic peptide from the N-terminal region (aa 1-100) of human IL-4; this sequence is largely conserved in mouse and rat, supporting the reported cross-reactivity.
- For Western blot, consider using reducing conditions and include appropriate positive controls such as stimulated T-cell or mast cell lysates. The predicted band is approximately 17 kDa, but glycosylation may cause slight shifts.
- In ELISA applications, the antibody can be explored as a capture or detection reagent; pairing with a matched monoclonal may improve sensitivity. Confirm performance in your specific sample matrix.
- When testing cross-reactivity in mouse or rat samples, validate with known positive and negative controls due to potential sequence variations.
