Biological Background: IL2 Function and Localization

IL2 (Interleukin-2), also known as T-cell growth factor (TCGF), is a secreted cytokine predominantly produced by activated CD4-positive helper T-cells and, to a lesser extent, by CD8-positive T-cells and natural killer cells.

It binds to a high-affinity trimeric IL-2 receptor (IL2RA/CD25, IL2RB/CD122, IL2RG/CD132) or a low-affinity dimeric receptor (IL2RB/CD122 and IL2RG/CD132), initiating JAK1 and JAK3 phosphorylation.

- The immunogen corresponds to amino acids 21-153 of human IL2, which represents the mature secreted form; thus the antibody may recognize the endogenous processed protein.
- For Western blot, consider using activated human T-cell lysates as a positive control; the predicted molecular weight is ~17 kDa, but glycosylation may result in a band between 15-20 kDa.
- For ELISA, a matched antibody pair may be required; validate the antibody’s performance as a capture or detection reagent in the relevant sample matrix.
- Cross-reactivity is reported for human; when working with other species, verify sequence homology and test experimentally.
