Biological Background: IL18 Function and Localization
- IL18 is a pro-inflammatory cytokine belonging to the IL-1 family, originally identified as interferon-gamma (IFN-gamma)-inducing factor. It is also known as iboctadekin, IL-1 gamma, and IGIF.
- It plays a central role in polarized T-helper 1 (Th1) and natural killer (NK) cell immune responses, promoting IFN-gamma synthesis in synergy with IL-12. Related references: PMID:10653850
- Upon binding to its receptors IL18R1 and IL18RAP, IL18 forms a ternary signaling complex that activates NF-kappa-B, leading to transcription of inflammatory mediator genes. Related references: PMID:14528293, PMID:25500532, PMID:37993714
- IL18 is involved in epithelial barrier repair and is released from cells via pyroptosis: its mature form translocates through gasdermin-D (GSDMD) pores into the extracellular space. Related references: PMID:33883744
- The protein localizes to the cytoplasm and cytosol, and is secreted. It is expressed in ovarian carcinoma cells but is undetectable in normal ovarian epithelial cells.
- IL18 is subject to alternative splicing, as indicated by its UniProt keyword annotation, and its activation is unexpectedly resistant to proteolytic cleavage by caspase-1 and caspase-4 in some contexts.
Experimental Guidance and Technical Tips
- This antibody was generated against full-length recombinant human pro-IL18 (amino acids 1-193). It is expected to recognize both the pro-form and the mature, cleaved form of IL18.
- For Western blotting, a band at approximately 22 kDa is predicted. Optimal working dilutions should be determined empirically.
- The antibody is validated for immunohistochemistry (paraffin) and ELISA. For IHC-P, appropriate antigen retrieval methods may be required; validate in the relevant tissue system.
- Due to cross-reactivity with mouse and rat, this antibody is a useful tool for comparative studies across these species; however, we recommend verifying reactivity in your specific assay conditions.
