Biological Background: Interleukin-10 Function and Localization
- Interleukin-10 (IL-10), also known as cytokine synthesis inhibitory factor (CSIF), is a secreted anti-inflammatory cytokine.
- IL-10 is a major immune regulator that limits excessive inflammation by acting on various immune cells, particularly antigen-presenting cells (APCs) such as macrophages and monocytes.
- It inhibits the release of multiple pro-inflammatory cytokines including GM-CSF, G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8, and TNF.
- IL-10 also interferes with antigen presentation by reducing expression of MHC class II and co-stimulatory molecules on APCs, thereby suppressing T cell activation (By similarity).
- Upon binding to its heterotetrameric receptor IL10RA/IL10RB, IL-10 triggers JAK1 and STAT2-mediated phosphorylation of STAT3, which translocates to the nucleus to drive anti-inflammatory gene expression.

- For ELISA, consider pairing with a matched detection antibody if developing a sandwich assay. Validate the antibody in the relevant mouse sample matrix to ensure performance.
- Since IL-10 is a secreted cytokine, analyzing cell culture supernatants or serum samples may be appropriate; note that endogenous levels can be low in resting cells and may require stimulation.
