Biological Background: Interleukin-10 Function and Localization

• Interleukin-10 (IL-10), also known as cytokine synthesis inhibitory factor (CSIF), is a secreted anti-inflammatory cytokine.
• IL-10 is a major immune regulator that limits excessive inflammation by acting on various immune cells, particularly antigen-presenting cells (APCs) such as macrophages and monocytes.
• It inhibits the release of multiple pro-inflammatory cytokines including GM-CSF, G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8, and TNF.
• IL-10 also interferes with antigen presentation by reducing expression of MHC class II and co-stimulatory molecules on APCs, thereby suppressing T cell activation (By similarity).
• Upon binding to its heterotetrameric receptor IL10RA/IL10RB, IL-10 triggers JAK1 and STAT2-mediated phosphorylation of STAT3, which translocates to the nucleus to drive anti-inflammatory gene expression.

• This rabbit polyclonal antibody was raised against a recombinant fusion protein encompassing amino acids 19-178 of mouse IL-10. The immunogen spans the mature cytokine sequence, which may contribute to recognition of native IL-10.
• For Western blotting (WB), mouse IL-10 has a predicted molecular weight of approximately 21 kDa; band detection in suitable samples may require optimization of protein loading and transfer conditions.
