Biological Background: IL-1 alpha Function and Localization
- Interleukin-1 alpha (IL-1α), also known as Hematopoietin-1, is encoded by the IL1A gene and belongs to the IL-1 family of cytokines.
- Constitutively expressed in nearly all resting non-hematopoietic cells, IL-1α functions as both an intracellular protein and a secreted cytokine that bridges innate and adaptive immunity. Related references: PMID:26439902
- Upon binding to IL1R1 and IL1RAP, it forms a high-affinity receptor complex, recruiting adaptors MYD88, IRAK1/4 and activating NF-κB, p38, p42/p44, and JNK signaling pathways. Related references: PMID:17507369, PMID:2950091, PMID:14687581
- Acts as an alarmin: passively released upon cell necrosis, alerting the host to tissue damage and triggering inflammation. Related references: PMID:15679580
- Intracellularly, IL-1α can directly sense DNA damage and serve as a signal for genotoxic stress without compromising cell membrane integrity. Related references: PMID:26439902
- Subcellularly localized to the nucleus, cytoplasm, and secreted into extracellular space, reflecting its dual intracellular/extracellular roles.
- Post-translational modifications include acetylation, glycosylation, lipidation (myristoylation), and phosphorylation, which may regulate its localization and activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region (aa 113-267) within the mature IL-1α protein, which may recognize both precursor and processed forms. For WB, consider a molecular weight around 31kDa (full-length precursor) and detect possible cleavage products.
- Given cross-reactivity with mouse and rat, this antibody can be utilized across species in comparative studies; however, we recommend validating in the relevant sample system.
- For IHC-P, optimize antigen retrieval and antibody dilution; since IL-1α is secreted and cytoplasmic/nuclear, assess staining patterns accordingly.
- For ELISA, consider pairing with a suitable detection antibody; this polyclonal pAb may serve as a capture or detection reagent after proper validation.
