Biological Background: IKKα (CHUK) Function and Localization
-
IKKα (CHUK) is a serine/threonine-protein kinase that acts as the catalytic subunit of the IKK complex, playing a central role in the canonical NF-kappa-B signaling pathway by phosphorylating IκB inhibitors, triggering their ubiquitination and proteasomal degradation.

It also participates in the non-canonical NF-kappa-B pathway, where the MAP3K14-activated IKKα homodimer phosphorylates NFKB2/p100, inducing its processing to p52 and formation of RelB-p52 complexes that regulate B-cell survival and lymphoid organogenesis.

IKKα exerts negative feedback control on both canonical and non-canonical NF-kappa-B pathways: it promotes assembly of the A20 ubiquitin-editing complex to limit inflammatory gene activation, and also phosphorylates and destabilizes MAP3K14/NIK.
Related references: PMID:21765415 -
Within the nucleus, IKKα phosphorylates CREBBP, enhancing its transcriptional and histone acetyltransferase activities, and also phosphorylates histone H3 at Ser-10, facilitating subsequent acetylation at Lys-14 by CREBBP to modulate chromatin accessibility at NF-kappa-B-responsive promoters.
Related references: PMID:17434128 PMID:12789342 -
IKKα phosphorylates FOXO3, potentially regulating its pro-apoptotic activity.
Related references: PMID:15084260 -
In the context of mitophagy, IKKα phosphorylates AMBRA1, promoting its interaction with ATG8 family proteins and stimulating mitophagic activity.
Related references: PMID:30217973 -
The protein is widely expressed and localizes to both the cytoplasm and nucleus, consistent with its dual roles in signaling and transcriptional regulation.
-
Key post-translational modifications include acetylation and ubiquitin conjugation, and IKKα contains an ATP-binding domain essential for its kinase function.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-100 at the N-terminus of human IKKα; this region is likely conserved in mouse, supporting the reported cross-reactivity. Validate in the relevant sample system before large-scale experiments.
- For Western blot, an expected band at approximately 85kDa should be observed. Optimize antibody dilution and blocking conditions for your specific lysates.
- In ELISA applications, consider using the immunogen peptide as a positive coating antigen to confirm antibody reactivity, and test cross-reactivity with mouse IKKα if needed.
- Note that multiple isoforms or post-translationally modified forms may produce additional bands; consult current literature for the specific model system.
