Biological Background: IGF2BP3 Function and Localization
- Also known as IMP3 and VICKZ3, belongs to the VICKZ family of mRNA-binding proteins.
- Functions as an RNA-binding protein that recruits target transcripts to cytoplasmic mRNA-protein complexes (mRNPs) for transport, transient storage, and translational regulation.
- Preferentially binds to N6-methyladenosine (m6A)-modified mRNAs, enhancing their stability; also stabilizes MYC mRNA by binding to the coding region instability determinant (CRD) in an m6A-dependent manner. Related references: PMID:29476152
- Binds to the 3'-UTR of CD44 mRNA, stabilizing it and promoting cell adhesion and invadopodia formation in cancer cells.
- Binds to beta-actin/ACTB and the 5'-UTR of insulin-like growth factor 2 (IGF2) mRNAs, regulating their expression.
- Localizes to the nucleus, cytoplasm, P-bodies, and stress granules, reflecting roles in mRNA processing, storage, and stress response.
- Expressed in fetal tissues (including liver, lung, kidney, thymus, placenta) and overexpressed in cervical, testicular, pancreatic, and renal cell carcinomas.
- Post-translational modifications include phosphorylation, isopeptide bond formation, and ubiquitination (based on UniProt keywords).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 300-579 of human IGF2BP3 (NP_006538.2), a region conserved in mouse, consistent with the reported cross-reactivity.
- For Western blotting, a band of approximately 64 kDa is expected; use appropriate positive and negative controls to validate specificity.
- For immunoprecipitation (IP), standard protocols may be used; verify capture efficiency by co-detection of known interacting mRNAs or proteins.
- For immunofluorescence/immunocytochemistry (IF/ICC) and immunohistochemistry (IHC-P), optimize fixation and permeabilization; nuclear and cytoplasmic staining may be observed.
- Consider using the antibody in ELISA with the immunogen peptide as a positive control; titration experiments are recommended.
