  • Insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1/IMP1) is an RNA-binding protein belonging to the VICKZ family, encoded by the IGF2BP1 gene. Also known as CRD-BP, ZBP1, or VICKZ1, it features multiple RNA-binding domains that recognize specific mRNA elements.
  • Core function: Recruits target transcripts into cytoplasmic mRNP complexes, enabling mRNA transport, temporary storage, and translational repression. Preferentially binds N6-methyladenosine (m6A)-modified mRNAs, enhancing their stability. Related references: PMID:29476152, PMID:32245947
  • Plays a key role in spatial regulation of ACTB/β-actin mRNA: binds the ‘zipcode’ element, transports the mRNP along the cytoskeleton, and releases the transcript upon phosphorylation, allowing local actin synthesis essential for cell polarity and migration.

  • For Western blotting, predicted full-length molecular weight is ~63 kDa; the datasheet lists 49 kDa/63 kDa. Use positive controls from cell lines known to express IGF2BP1 (e.g., embryonic or cancer lines) and validate band identity.
  • In IHC-P and IF/ICC, IGF2BP1 localizes to both nucleus and cytoplasm, with potential enrichment in cell protrusions. Optimize permeabilization and include isotype controls.
