Biological Background: IFNAR2 Function and Localization

  • IFNAR2 (interferon alpha/beta receptor 2), also known as IFNABR, IFNARB, or IFN-alpha-REC, encodes the type I interferon receptor beta chain. Multiple isoforms are generated by alternative splicing; notably, isoform 3 is a secreted form detected in urine (PubMed:7759950, PubMed:8181059).   • Together with IFNAR1, IFNAR2 forms the functional heterodimeric receptor for type I interferons (IFN-α, -β, -ε, -ω, -κ) (PubMed:10049744, PubMed:10556041, PubMed:21854986).   • For Western blotting, a band at ~58 kDa is predicted; however, due to N-linked glycosylation and potential proteolytic processing, additional bands or a higher apparent molecular weight may be observed. Validate with positive (e.g., IFN-treated cells) and negative controls.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), membrane staining is anticipated. For tissues or cells expressing the secreted isoform, cytoplasmic or extracellular matrix signal may be detected; appropriate fixation and permeabilization protocols should be tested.
  • For ELISA, this polyclonal antibody can be used as a detection reagent when paired with a suitable monoclonal capture antibody; cross-reactivity with mouse and rat IFNAR2 should be verified in the assay matrix.
  • As a polyclonal antibody, lot-to-lot consistency should be monitored, and titration experiments are recommended for new applications.
