Biological Background: IFN gamma Function and Localization
-
Interferon gamma (IFN-γ), encoded by the IFNG gene, is a type II interferon and a key secreted cytokine belonging to the interferon family. It is also known as immune interferon (If2f; IFNG2) and is primarily released from activated T lymphocytes.
-
It plays a critical role in antimicrobial, antiviral, and antitumor immune responses by activating effector immune cells and enhancing antigen presentation. Related references: PMID:16914093, PMID:8666937
-
IFN-γ signals through the JAK-STAT pathway upon binding to its receptor IFNGR1, leading to activation of STAT1 and transcription of IFN-γ-regulated genes, many of which are transcription factors like IRF1 that further amplify the immune response. Related references: PMID:16914093, PMID:8349687
-
It enhances antigen presentation by inducing the replacement of constitutive proteasome subunits with immunoproteasome subunits, thereby increasing the quantity, quality, and repertoire of peptides for MHC class I loading. Related references: PMID:8666937, PMID:8163024
-
IFN-γ also upregulates MHC class II complexes on the cell surface by promoting expression of several key molecules such as cathepsins B/CTSB, H/CTSH, and L/CTSL. Related references: PMID:7729559
-
It increases the efficiency of peptide generation by inducing the activator PA28, which associates with the proteasome and alters its proteolytic cleavage preference. Related references: PMID:11112687
-
Post-translationally, IFN-γ is a glycoprotein with a signal peptide, and it undergoes cleavage on pairs of basic residues and contains a pyrrolidone carboxylic acid modification.
-
Under homeostatic conditions, IFN-γ participates in the regulation of hematopoietic stem cells, influencing their development, quiescence, and differentiation.
Experimental Guidance and Technical Tips
- For Western blotting (WB), the predicted molecular weight of IFN-γ is approximately 18 kDa; consider using reducing conditions and validate in the relevant sample system (e.g., stimulated T-cell lysates or recombinant protein controls).
- In ELISA applications, this antibody may be used as a detection or capture antibody; pairing with a validated matched antibody pair is recommended and requires individual optimization.
- Since the immunogen is a recombinant protein covering human and rat IFN-γ, cross-reactivity with other species should be verified experimentally if needed.
