Biological Background: IFN-beta Function and Localization
- Interferon beta (IFN-beta, IFNB1) is a type I interferon cytokine encoded by the Ifnb1 gene in mice, playing a central role in innate immune responses against viral and bacterial infections (PubMed:10708458, 23872679). Related references: PMID:10708458, PMID:23872679
- It signals through the heterodimeric receptor composed of IFNAR1 and IFNAR2, activating the canonical JAK-STAT signaling cascade, which leads to transcriptional regulation of interferon-stimulated genes (ISGs) (PubMed:23872679). Related references: PMID:23872679
- IFN-beta exhibits potent antiviral and antibacterial activities, and modulates immune cell development such as B-cell maturation and myelopoiesis, as well as LPS-induced tumor necrosis factor production (PubMed:10708458, 14597717). Related references: PMID:10708458, PMID:14597717
- Beyond immunomodulation, IFN-beta maintains neuronal homeostasis by promoting autophagy and clearance of alpha-synuclein, thereby protecting dopaminergic neurons from degeneration (PubMed:26451483). Related references: PMID:26451483
- Subcellular localization is primarily secreted, consistent with its role as an extracellular signaling molecule.
- Key structural features include a signal peptide, disulfide bonds, and glycosylation, which contribute to its stability and bioactivity.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein covering amino acids 1-182 of mouse IFN-beta (NP_034640.1), representing the full-length mature protein. This rabbit monoclonal antibody is expected to recognize the native mouse IFN-beta protein.
- For Western blot (WB), a band migrating around 22 kDa is expected, but due to glycosylation and other post-translational modifications, the observed molecular weight may be higher. Validate in relevant mouse samples, such as stimulated immune cell lysates.
- For ELISA, the antibody may be tested in both direct and sandwich formats. Pairing with another IFN-beta antibody or using as a detection antibody could be considered. Confirm cross-reactivity and specificity with mouse IFN-beta and lack of cross-reactivity with other type I IFNs.
