Biological Background: C6 Function and Localization
- As a core component of the membrane attack complex (MAC), complement C6 contributes to the formation of transmembrane pores that disrupt target cell membranes, ultimately leading to cell lysis and pathogen clearance. Related references: PMID:22267737 / PMID:22832194 / PMID:26841837
- C6 associates with complement C5b on the outer leaflet of target cell membranes, facilitating MAC assembly by reducing the energy required for membrane bending and pore formation. Related references: PMID:30552328 / PMID:32569291
- MAC formation is triggered by proteolytic cleavage of C5 into C5b, which occurs in response to activation of the classical, alternative, lectin, and GZMK complement pathways. Related references: PMID:30552328 / PMID:39914456 / PMID:39814882
- C6 is a secreted protein that localizes to the target cell membrane upon complement activation, inserting into the lipid bilayer as part of the MAC.
- The protein features multiple structural domains, including EGF-like and sushi (SCR) repeats, and undergoes post-translational modifications such as glycosylation and disulfide bond formation.
- As an effector of innate immunity, C6 plays a critical role in defending against pathogens by directly lysing target cells and promoting downstream immune signaling.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning the C-terminal region (amino acids 586–934) of human C6. This design may enhance specificity for full-length C6; however, cross-reactivity with other complement components should be assessed in relevant assays.
- For Western blot (WB), a band at approximately 105 kDa is expected. It is advisable to use pre-stained molecular weight markers and optimize transfer conditions for high-molecular-weight proteins.
- In ELISA applications, consider coating with recombinant C6 or the immunogen fragment as a positive control, and include appropriate negative controls to confirm signal specificity.
