Biological Background: Advanced Glycation End Product-Specific Receptor (AGER/RAGE) Function and Localization
- Recognizes a broad range of endogenous stress signals including advanced glycation end products (AGEs), S100 proteins, HMGB1, amyloid-beta, nucleic acids, and histones. Related references: PMID:27572515, PMID:28515150, PMID:34743181
- Upon ligand binding, initiates pro-inflammatory signaling via the TIRAP/MYD88 pathway, leading to NF-kappa-B activation and induction of cytokines such as IL6, IL8, and TNFalpha. Related references: PMID:21829704, PMID:33436632
- Mediates transcytosis of amyloid-beta peptide across the blood-brain barrier and contributes to neurodegenerative processes. Related references: PMID:19906677
- Promotes phagocytosis by non-professional phagocytes, facilitated by binding to RNA, DNA, HMGB1, and histones. Related references: PMID:35974093
- Translocates to the nucleus upon DNA damage and enhances DNA repair through interaction with the MRN complex and MRE11. Related references: PMID:33918759
- Highly expressed in endothelial cells and upregulated in inflammatory conditions and in pre-term labor and preeclampsia placentas. Related references: PMID:33918759
- Involved in endothelial dysfunction and reactive oxygen species (ROS) production. Related references: PMID:25401185
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 24-344) of human AGER; the antibody may recognize both membrane-bound and soluble forms of the receptor.
- For Western blot, the predicted molecular weight is approximately 43 kDa; use appropriate positive controls such as endothelial or lung tissue lysates.
- Due to reported cross-reactivity with mouse and rat, this antibody may be suitable for studies in rodent models, but validation in the specific sample system is recommended.
- For ELISA applications, consider pairing with a suitable detection antibody and optimizing assay conditions.
