Biological Background: HTRA3 Function and Localization
- Serine protease that cleaves beta-casein (CSN2) and extracellular matrix proteoglycans including decorin (DCN), biglycan (BGN), and fibronectin (FN1). (By similarity)
- Inhibits TGF-beta family signaling possibly indirectly through degradation of these ECM proteoglycans. (By similarity)
- May act as a tumor suppressor. Related references: PMID:21321049 PMID:22229724
- Negatively regulates trophoblast invasion during placental development in vitro, and may be involved in placental development in vivo. Related references: PMID:21321049 PMID:22229724
- May have a role in ovarian development, granulosa cell differentiation, and luteinization. Related references: PMID:21321049 PMID:22229724
- Widely expressed, with highest levels in both adult and fetal heart, ovary, uterus, placenta, and bladder. In the endometrium, expressed in epithelial glands and the stroma. Also present in leukocytes. Isoform 1 is predominant in heart and skeletal muscle, whereas isoform 2 is predominant in placenta and kidney.
- Secreted protein belonging to the high-temperature requirement A (HTRA) family; alternative splicing yields multiple isoforms.
- Keywords indicative of structural and functional features: 3D-structure, alternative splicing, disulfide bond, hydrolase, protease, secreted, serine protease, signal.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 334-453, a region likely conserved across isoforms; verify isoform recognition if differential expression is relevant to your study.
- For Western blotting, the predicted molecular weight is approximately 49 kDa (full-length), but migration may shift due to post-translational modifications; validate in appropriate tissue lysates with loading controls.
- For ELISA, optimize coating concentration and detection antibody dilution; recombinant HTRA3 protein can serve as a standard.
- Cross-reactivity with mouse and rat has been confirmed, making this antibody suitable for rodent model studies; validate in the specific strain and tissue of interest.
- Given HTRA3's involvement in reproductive biology and tumor suppression, recommended sample types include placental, ovarian, and tumor cell line extracts.
