Biological Background: HSD17B2 Function and Localization
- HSD17B2 is a 17-beta-hydroxysteroid dehydrogenase that catalyzes the NAD-dependent oxidation of the highly active 17beta-hydroxysteroids, such as estradiol (E2), testosterone (T), and dihydrotestosterone (DHT), to their less active forms, thereby regulating biological potency.
- It converts estradiol to estrone, testosterone to androstenedione, and dihydrotestosterone to 5alpha-androstan-3,17-dione. This enzyme also exhibits 20-alpha-hydroxysteroid dehydrogenase activity.
- HSD17B2 belongs to the short-chain dehydrogenase/reductase (SDR) family, specifically member 9C2 (SDR9C2).
- The protein localizes to the endoplasmic reticulum membrane, consistent with its role in steroid metabolism.
- Tissue expression is prominently reported in the placenta, indicating a role in modulating hormone levels at the maternal-fetal interface.
- According to UniProt, the calculated molecular weight is approximately 42.8 kDa, and the protein features a signal-anchor and transmembrane helix, anchoring it to the membrane.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 100–200 of human HSD17B2 (NP_002144.1). When using this antibody in WB, consider that it may recognize isoforms or fragments containing this region.
- For Western blotting, the expected band is around 43 kDa. Validate sensitivity and specificity using positive control lysates, such as placental tissue or cell lines expressing HSD17B2.
- This antibody is qualified for ELISA. Optimal coating antigen and antibody dilutions should be determined experimentally by the end user.
- Due to cross-reactivity with mouse and rat, this antibody may be suitable for studies across these species. However, we recommend confirming reactivity with the specific sample system in preliminary experiments.
